Whole genome sequencing of 160 marine fish for the FORESEA project, 2022
Genetic diversity is often considered a key factor in a species’ ability to adapt to environmental changes and our aim was to understand the biological, ecological, and biogeographic factors that explain differences in genetic diversity among marine fish. Therefore, whole genomes of 160 individual marine fish were sequenced to a minimum depth of 15×, enabling near-complete representation of each diploid genome.

- Genetic diversity is often considered a key factor in a species’ ability to adapt to environmental changes. However, there is variability in genetic diversity among species, and the natural determinants of this variability remain poorly understood, particularly within certain taxonomic groups. The objective of this study, conducted as part of the FORESEA project, was to understand the biological, ecological, and biogeographic factors that explain differences in genetic diversity among marine fish. Therefore, we collected a total of 160 marine fish individuals, encompassing 100 different species. Sampling was conducted in two geographic regions: the Gulf of Lion in the Mediterranean and the Bay of Biscay in the Atlantic (occasionally extending to the English Channel). Our goal was to get one individual per species to represent as many species as possible in both regions. Tissue samples were preserved in 95% ethanol prior to the extraction of total genomic DNA. Subsequently, genomic libraries were prepared by Integragen, and two sequencing runs were performed using the Illumina NovaSeq 6000 platform. Whole genomes of each individual were sequenced to a minimum depth of 15×, enabling near-complete representation of each diploid genome.
